Screening of Microorganism Producing Chitosanase Using Trypan Blue Containing Medium and Characterization

Abstract
We were isolated one strain producing chitosanase from Doen-jang, a Korea traditional fermented food. When the chitosan was used as substrate, the strain formed clear zone by its chitosanase activity on Trypan Blue containing plate. Based on theanalysis of 16S rRNA gene sequence from the strain, the isolated strain was identified with Bacillus methylotrophicus, therefore we named the strain Bacillus methylotrophicus CH1.The B. methylotrophicus CH1 produced chitosanase maximally after 48h cultivation, and the enzyme from B. methylotrophicus CH1 was optimal at pH 5.0 and 50¡ÆC. In the investigation of substrate specificity for 43, 67, 223, 444, and 789 kDa chitosan, chitosanase from B. methylotrophicus CH1was showed the highest activity for 43 kDa chitosan. In the analysis of the reaction products of chitosanase from B. methylotrophicus by thin-layer chromatography, the various chitosan oligosaccharides were detected as main products from 43 kDa chitosan.
